Continuous perfusion processes rely on the accurate delivery of large medium volumes over extended cultivation periods. In practice, mechanical variability, backpressure effects, and limited measurement resolution often undermine rate-based feeding strategies, leading to cumulative dosing errors.

In collaboration with Safi Biotherapeutics, Securecell has developed a practical control strategy that addresses these challenges by applying gravimetric corrections to the feed delivery. Rather than the instantaneous feed rate, these corrections are based on the cumulative amount of medium delivered. By regulating the integrated mass of feed over time, the strategy compensates for pump drift and measurement noise and maintains accurate overall dosing.

Applied to perfusion processes, the approach enabled stable and reproducible operation under variable hydraulic conditions and common operational disturbances. The results highlight gravimetric control as a practical and robust foundation for perfusion process development in complex R&D environments.

In the webinar “Implementing Gravimetric Feed Control in Perfusion-Based Cell Culture Processes,” a successful gravimetric control strategy will be presented and its performance discussed.
